Abstract

Three methods for extraction (infusion, maceration and maceration with ultrasound) for the preparation of ethanol extracts from seed and quantification of coumarin in Tonka bean seed, the active ingredient associated with medical applications of this plant were evaluated. The evaluation showed that the ultrasonic maceration gave the best results in terms of performance and time of the extraction. A method by High-performance liquid chromatography with ultraviolet detection (HPLC-UV) for the detection and quantification of coumarin in ethanol extracts of the seed of D. odorata is described. The separation was performed in isocratic mode with mobile phase 45% acetonitrile and 55% water, on a Resolve C18 column of 5 μm. The mobile phase flow was 1.00 ml x min-1, with detection at 275 nm. The calibration and validation parameters (LOD = 0.07 mg x g-1 seed, LOQ = 108 mg x g1 seed in a linear range between LOQ to LOQ x30; % RSD <5 for all calibration points proved a reproducible and sensitive method for the determination of coumarin in this matrix.
