Abstract

SEGURA-VALDEZ, María de Lourdes et al. Observations on the nuclear structure of onion (Allium cepa L.) meristematic root cells by atomic force microscopy. TIP [online]. 2006, vol.9, n.1, pp.30-33. ISSN 1405-888X.

The in situ structure of the cell has been extensively studied by light and electron microscopy. Recently, atomic force microscopy has been also used to this aim. In this study, atomic force microscopy observations on the nuclear structure are presented from semi-thin sections of material prepared for standard methods of electron microscopy. In the onion root tip cell nuclei, clumps of compact chromatin, nucleoplasm space, nuclear pores and nucleoli are observed. Those images well correspond to those observed by bright field, although at higher resolution.

Keywords : Onion; chromatin; atomic force microscopy (AFM); nucleus; nucleolus.
