Abstract

Five native isolates of the fungus Pochonia chlamydosporia, a biological control agent of root-knot and cyst-forming nematodes, were collected from Nacobbus aberrans sensu stricto infested plots in different localities in the states of Mexico, Morelos, Puebla and Tlaxcala (Mexico). Isolation and culturing methods are described for isolates SMB3A, SC1, SMB3, SM4, MHCH, and evaluation of the parasitism of nematode eggs and rhizosphere colonization by the fungus. Molecular characterization of the isolates was made using β-tubulin-PCR, ERIC-PCR (enterobacterial repetitive intergenic consensus sequence), ITS-PCR (internally transcribed spacer regions), RFLP-PCR (restriction fragment length polymorphism), RAPD-PCR (random amplified polymorphic DNA) and sequencing of DNA fragments. More than 60% of nematode eggs were parasitized by the Mexican isolates. All isolates colonised the complete extent of the rhizosphere. All isolates were identified using species-specific primers (β-tubulin-PCR) as belonging to Pochonia chlamydosporia var. chlamydosporia.
