VELEZ TORRES, Marcelina et al. Obtaining haploid plants from miahuateco chili pepper (Capsicum annuum L.). Rev. Mex. Cienc. Agríc [online]. 2010, vol.1, n.2, pp.189-201. ISSN 2007-0934.
Haploid plant regeneration is an important tool in breeding programs and genetics studies, since it helps obtain pure lines faster than conventional methods by the duplication of haploid plants. The aim of this study was to establish a methodology to regenerate haploid Miahuateco chili pepper plants (Capsicum annuum L.). Anthers were grown on Murashige and Skoog (1962); Chu et al. (1975) basal media, supplemented with 6-furfurylaminopurine (0.1-1 mg L-1), naphthaleneacetic acid (0.1 mg L-1), indolacetic acid (1 mg L-1), and 2-4 dichlorophenoxyacetic acid (1 mg L-1). Embryogenesis was induced in 2.23% of anthers grown in a combination of 6-furfurylaminopurine with 2-4 dichlorophenoxyacetic acid (1 mg L-1, of each), or indolacetic acid with 6-furfurylaminopurine (0.1 mg L-1 of each). Chromosome analysis of regenerated plants showed that they were haploids with a chromosome number of 2n= x= 12.
