Abstract

The ability of Pseudomonas sp. to associate with and penetrate potato seedling roots cv. Alpha propagated by tissue culture was evaluated. 38-day old potato seedlings were inoculated with 100 μL of a bacterial suspension containing 108 CFU mL-1 of culture medium. A biofilm was observed surrounding the roots after seven days of incubation. The initial concentration of bacterial cells inside root plants was 1.53 X 107 CFU g-1 of plant tissue. In the fifth week of the incubation period the concentration was 1.92 X 105 CFU g-1 of plant tissue, which was similar to that obtained in the support medium. The digestion patterns of the ribosomal gene 16S of the DNAr showed that, with this kind of inoculation, Pseudomonas sp. can be established in an endophytic way and guarantee that the seedlings will associate only with bacteria of interest. In further investigations, aspects such as promotion of plant growth and degradation of certain agrochemicals will be evaluated.
