El frijol es un producto básico en la dieta de la población, por su alto contenido de proteínas y elementos minerales esenciales. Sin embargo, muchas variedades de frijol muestran oscurecimiento del grano durante el almacenamiento, lo que provoca depreciación debido a la poca aceptación de los consumidores que no están dispuestos a pagar un precio normal e induce pérdidas económicas para los productores y comercializadores. El oscurecimiento del grano de frijol es un proceso complejo que involucra la acción de diversas enzimas, entre las que se encuentran las fenol oxidasas. Los marcadores moleculares de ADN asociados al oscurecimiento han tenido un comportamiento heterogéneo, siendo útiles para algunos tipos de frijol, como el pinto, pero no para otros, como flor de mayo o flor de junio. El objetivo fue identificar genes de fenol oxidasas asociados al oscurecimiento de la testa del grano de distintos tipos de frijol, cuyos niveles de expresión sean útiles como marcadores para identificar genotipos de oscurecimiento lento. Se estudiaron los perfiles de expresión de ocho fenol oxidasas mediante RT-PCR en semillas de cuatro variedades de frijol pinto, dos flor de junio y dos flor de mayo con diferente comportamiento al oscurecimiento de la testa. Se demostró que la acumulación de transcritos de los genes difenol oxidasa lacasa 1, difenol oxidasa lacasa 2, difenol oxidasa lacasa 3 y polifenol oxidasa 2 están asociados con en el oscurecimiento lento de la semilla de frijol.]]></p></abstract>
<abstract abstract-type="short" xml:lang="en"><p><![CDATA[Summary The grown of beans (Phaseolus vulgaris L.) is one of the main activities of the peasant economy and an important generator of income and rural employment. Beans are a staple in people&#8217;s diets because of their high protein and essential mineral content. However, many bean varieties show darkening of the bean during storage, which causes depreciation due to poor acceptance by consumers who are unwilling to pay a normal price and induces economic losses for farmers and marketers. Bean darkening is a complex process involving the action of several enzymes, including phenol oxidases. DNA molecular markers associated with darkening have had a heterogeneous behavior, being useful for some bean types, such as pinto, but not for others, such as May flower or June flower. The objective was to identify phenol oxidase genes associated with darkening of the bean testa of different bean types, whose expression levels are useful as markers to identify slow darkening genotypes. The expression profiles of eight phenol oxidases were studied by RT-PCR in seeds of four pinto bean varieties, two June flower and two May flower with different testa darkening behavior. It was shown that the accumulation of transcripts of diphenol oxidase laccase 1, diphenol oxidase laccase 2, diphenol oxidase laccase 3 and polyphenol oxidase 2 genes are associated with slow bean seed darkening.]]></p></abstract>
