<self-uri xlink:href="http://www.scielo.org.mx/scielo.php?script=sci_arttext&amp;pid=S0188-62662016000400056&amp;lng=en&amp;nrm=iso"></self-uri><self-uri xlink:href="http://www.scielo.org.mx/scielo.php?script=sci_abstract&amp;pid=S0188-62662016000400056&amp;lng=en&amp;nrm=iso"></self-uri><self-uri xlink:href="http://www.scielo.org.mx/scielo.php?script=sci_pdf&amp;pid=S0188-62662016000400056&amp;lng=en&amp;nrm=iso"></self-uri><abstract abstract-type="short" xml:lang="es"><p><![CDATA[Resumen Los estudios de variabilidad genética han reportado inconsistencias de resultados entre poblaciones, en gran medida, debido a que los métodos de extracción de ácido desoxirribonucleico (DNA, por sus siglas en inglés) y las técnicas de genotipificación son altamente variables entre ellas, lo que conduce a la asignación errónea de genotipos. El objetivo de este estudio es comparar distintas técnicas de extracción de DNA y de genotipificación de polimorfismos. Para llevar a cabo el estudio, se analizó el DNA de 10 muestras sanguíneas correspondientes a individuos mestizos mexicanos, y se purificó por los métodos de fenol-cloroformo-alcohol isoamílico (FCI), gradiente de sales (GS), gradiente de sacarosa (GSC), DNAzol® y DNeasy Blood &amp; Tissue Kit ®. Se genotipificaron los polimorfismos GSTT1 *0 y GSTM1*0 por reacción en cadena de la polimerasa (PCR, por sus siglas en inglés) múltiple, CYP1A1*2C por RFLP&#8217;s y AhR Arg554Lys por PCR tiempo real. Los resultados mostraron diferencias significativas (p &lt; 0.001) en cantidad, pureza e integridad entre los distintos métodos. En los análisis moleculares se observó que el método de FCI presenta inhibidores de la reacción de PCR, ya que no fue posible amplificar los fragmentos por PCR múltiple y PCR punto final, aunque sí hubo amplificación en el PCR tiempo real. Los métodos GS y GSC amplificaron todas las muestras en las tres modalidades de PCR y mostraron resultados concordantes, mientras que solo el 80% de las muestras extraídas mediante DNAzol ® y DNeasy ® amplificaron, y los resultados no fueron concordantes para DNAzol ® (50%) y DNeasy ® (80%) en el análisis de PCR - RFLP&#8217;s. Los métodos de extracción GS y GSC mostraron mayor recuperación de DNA, con parámetros de calidad e integridad óptimos para los análisis moleculares por PCR.]]></p></abstract>
<abstract abstract-type="short" xml:lang="en"><p><![CDATA[Abstract Genetic variability studies have presented inconsistencies between populations, mainly because methods of deoxyribonucleic acid (DNA) extraction and genotyping techniques show high variability between them, leading to an erroneous assignment of genotypes. The objective of this study is compare different techniques for DNA extraction and genotyping of polymorphisms. To perform this study, DNA from 10 blood samples corresponding to mestizo Mexicans was analyzed and purified by methods of phenol-chloroform-isoamyl alcohol (PCA), salting out gradient (SG), sucrose gradient (SCG), DNAzol ® method and DNeasy Blood &amp; Tissue Kit ®. GSTT1*0 and GSTM1*0 polymorphisms were genotyped by multiplex PCR, CYP1A1*2C by PCR-RFLP&#8217;s assay and AhR Arg554Lys by real-time PCR. Results shown that amount, purity and integrity of DNA were evaluated. Different methods results showed significant differences (p &lt; 0.001). Molecular analyses showed that PCA method presents inhibitors for PCR reaction because it was not possible to amplify fragments in multiplex PCR and endpoint. Although, there was amplification in real time PCR. SG and SCG methods amplified all samples in three types of PCR and the results were concordant between them. While in PCR - RFLP analysis, samples extracted by DNAzol ® and DNeasy ® amplified only 80% of samples with no concordant results (50% for DNAzol ® and 80% for DNeasy ®). SG extraction methods and SCG showed higher DNA recovery, with optimal quality parameters for molecular analyses by PCR.]]></p></abstract>
