La línea celular BUVEC E6E7 de origen bovino se utilizó en ensayos previos para intentar reproducirla sin éxito. En parte debido a la carencia de información de sus propiedades de crecimiento in vitro. Con este propósito se plantearon experimentos que procuraran información útil en la aplicación del procedimiento. Las células se obtuvieron del Instituto de Neurobiología/UNAM, fueron mantenidas en Medio F-12 (HAM) suplementado con suero fetal bovino y NaHCO3 e incubadas a 37 °C en atmósfera de 5% CO2 en aire saturado de humedad. El primer ensayo en placas de 24 pozos, se diseñó para determinar la dosis mínima para empezar un cultivo, resultando el valor de 2,500 células/pozo; en el segundo se identificó la densidad mínima de siembra a partir de cultivos con 1, 3, 9 y 27 células/mm2, cosechados con solución Tripsina-EDTA al alcanzar &gt;95 % de confluencia. Los valores promedio alcanzados fueron: 8.0 x 104, 9.3 x 104, 6.3 x 104 y 6.5 x 104 células totales, respectivamente. Para evaluar la cinética del desarrollo celular, se utilizaron por triplicado cajas de Petri de 35 mm Ø iniciando con una densidad de 3.0 células/mm2 e intervalos de medición cada 8 h; agrupadas en dos series. Una con cambio del medio cada 48 h y la otra sin cambio de medio. El mayor crecimiento observado en ambas fue a las 152 h, con una densidad promedio de 2,505 células/mm2 en la primera serie y 1,708 células/mm2 en la segunda. El tiempo de duplicación estimado fue de 11.73 y 12.6 h, respectivamente. La línea BUVEC E6E7 de origen endotelial expresa un vigor de crecimiento inusual.]]></p></abstract>
<abstract abstract-type="short" xml:lang="en"><p><![CDATA[Abstract: It has been suggested that rickettsia Anaplasma marginale might interact with endothelial cells in the bovine host. BUVEC E6E7 endothelial cell line has been utilized to grow Anaplasma marginale with no success, partially because in vitro growth characteristics were unknown. Therefore, several experiments were assayed in an attempt to retrieve useful information to define them. Growing culture cells were obtained from UNAM&#8217;s Neurobiology Institute, maintained with fetal calf serum and NaHCO3 supplemented F-12 (HAM) medium and incubated at 37 °C on 5% CO2-air humid mixture atmosphere. First two assays used 24 well plates were focused on determination of a minimum initial cell concentration, which was 2,500 cells/well and cell density, whose experiment was set with 1, 3, 9 and 27 cells/mm2. Cells were harvested with an EDTA-Trypsin solution when confluence was reached. Average final concentration of 8.0 x 104, 9.3 x 104, 6.3 x 104 &amp; 6.5 x 104 cells/well was respectively found. 35 mm Ø sterile Petri dishes were used for kinetics studies. Cultures were set in triplicate with a seed density 3 cell/mm2 divided into two series and sampling every 8 h. A 48 h culture media replacement interval applied for the first series and none for the second. Maximum growth was observed in both series at 152 h with 11.73 and 12.6 h doubling time in culture, respectively. It was confirmed that BUVEC E6E7 endothelial cell line expresses unusual growth strength.]]></p></abstract>
