Some cyanobacterial genera have the potential to produce secondary metabolites that are highly toxic to humans, livestock and wildlife. Reliable methods for the detection of cyanobacterial toxins with high specificity and low limits of detection are in high demand. Here we test a relatively new hybrid high resolution accurate mass quadrupole time-of-flight mass spectrometry platform (TripleTOF) for the analysis of cyanobacterial toxins in freshwater samples. We developed a new method that allows the quantitative analysis of four commonly observed microcystin congeners (LR, LA, YR, and RR) and anatoxin-a in a 6-min LC run without solid-phase enrichment. Limits of detection for the microcystin congeners (LR, LA, YR, and RR) and anatoxin-a were &lt;5 ng/L (200-fold lower than the guideline value of 1 µg/L as maximum allowable concentration of MC-LR in drinking water). The method was applied for screening freshwaters in the Pacific Northwest during the bloom and post-bloom periods. The use of high resolution mass spectrometry and concomitant high sensitivity detection of specific fragment ions with high mass accuracy provides an integrated approach for the simultaneous identification and quantification of cyanobacterial toxins. The method is sensitive enough for detecting the toxins in single Microcystis colonies.]]></p></abstract>
<abstract abstract-type="short" xml:lang="es"><p><![CDATA[Resumen: La presencia de algas nocivas en cuerpos de agua dulce rica en nutrientes se ha visto incrementada a nivel mundial, incluyendo la zona del noroeste del Pacífico. Algunas cianobacterias tienen el potencial de producir metabolitos secundarios altamente tóxicos para los humanos, la ganadería y la vida silvestre. Así pues, existe gran demanda de métodos específicos, confiables y con bajos límites de detección para el análisis de toxinas provenientes de cianobacterias. En el presente estudio, se ha utilizado una plataforma híbrida relativamente nueva basada en un analizador de alta resolución tipo triple cuadrupolo-Tiempo de Vuelo (TripleTOF) para el análisis de cianotoxinas en muestras de agua dulce. Se ha puesto a punto un procedimiento que permite el análisis cuantitativo de cuatro microcistinas observadas comúnmente (LR, LA, YR, y RR), así como una anatoxina-a en una separación cromatográfica de 6 minutos sin necesidad de enriquecimiento en fase sólida. Los límites de detección de las microcistinas y anatoxina-a fueron &lt;5 ng/L (200 veces inferior al valor de referencia de 1 µg/L, máximo permitido en agua potable para MC-LR). El procedimiento fue aplicado para detección en cuerpos de agua dulce del noroeste del pacifico durante el periodo de auge y post-auge cianobacteriano. El uso de espectrometría de masas de alta resolución, la alta sensibilidad de detección de iones fragmento específicos aunados a la alta exactitud de masa, proporcionan un enfoque integral para la detección y cuantificación simultanea de toxinas provenientes de cianobacterianas. Este procedimiento tiene la sensibilidad suficiente para detectar toxinas en una solo colonia de Microcystis.]]></p></abstract>
