El conocimiento de la interacción planta-bacteria puede proporcionar herramientas que ayuden a controlar esta enfermedad. Los perfiles de expresión de genes del tomate relacionados con defensa y de genes de patogenicidad de la bacteria, podrían permitir identificar genes esenciales para el bloqueo o desarrollo de la marchitez. La técnica de RT-qPCR permite cuantificar los niveles de expresión genética; sin embargo, es importante evaluar la eficiencia y el rango dinámico de los oligonucleótidos a utilizar, para descartar la posibilidad de que las diferencias observadas estén influenciadas por aspectos técnicos y no sólo por causas biológicas. Con el objetivo de identificar pares de oligonucleótidos que bajo las condiciones experimentales utilizadas se desempeñen adecuadamente, en este trabajo se evaluó el desempeño de 40 pares de oligonucleótidos, tanto previamente reportados como sintetizados de novo, 23 pares están asociados a 16 genes del tomate, y 17 pares asociados a 10 genes de R. solanacearum. Los resultados mostraron que oligonucleótidos con un desempeño adecuado en un laboratorio no siempre se comportan adecuadamente bajo las condiciones utilizadas en otro laboratorio, lo que subraya la importancia de revisar las secuencias y evaluar cada par de oligonucleótidos con los reactivos y equipo con que se pretende evaluar los niveles de expresión genética, independientemente de los reportes previos sobre el desempeño de tales oligonucleótidos.]]></p></abstract>
<abstract abstract-type="short" xml:lang="en"><p><![CDATA[Abstract: Bacterial wilt caused by Ralstonia solanacearum affects the tomato crop in several parts of México. Knowledge of plant-bacteria interaction might provide tools to control this disease. The expression profiles of genes related with defense in tomato and pathogenicity in R. solanacearum, could allow the identification of essential genes for blocking wilt development. The RT-qPCR technique is suitable to quantify the levels of gene expression. However, it is important to evaluate the efficiency and dynamic range of oligonucleotides to be used in order to rule out the possibility that the differences observed are influenced by technical issues and not only by biological causes. With the aim of identifying oligonucleotide pairs that can perform properly under the experimental conditions used, in this study, the performance of 40 oligonucleotides pairs, both previously reported and de novo synthesized, were evaluated; 23 of these pairs are associated to 16 genes of tomato and 17 are associated to 10 genes of R. solanacearum. Results showed that those oligonucleotides with adequate performance in one laboratory, not always perform adequately under the conditions prevalent in other laboratory, which underlines the importance of reviewing sequences and evaluating each pair of oligonucleotides with the reagents and equipment with which the gene expression levels are going to be evaluated, regardless of previous reports on the performance of such oligonucleotides.]]></p></abstract>
