Simultaneous purification conditions of phytosterols and squalene were optimized for its analysis in soybean oil deodorizer distillate, a by-product of the oil refining industry. The combined effect of the eluent volume, the amount of silica gel and amount of compounds in the sample were studied using a complete factorial arrange 23 in a central compound design. The purification of phytosterols was mainly influenced by the first two factors (p<0.05) but not by the third factor. The analytical method was validated, obtaining precision and accuracy that were acceptable in reproducibility conditions. The concentrations of total phytosterols, free and esterified phytosterols and squalene, were: 17.19, 15.16, 2.03, y 2.17 % p/p respectively, making this by-product a potential source of said compounds for the food, pharmaceutical and cosmetic industry.
