A simple, fast and reliable analytical method for the quantification of iohexol in human serum by HPLC was developed. Iohexol extraction was done by protein precipitation with perchloric acid (5%, v/v in water). Sulfapyridine was used as internal standard (IS). The mixture was separated using a C18 column (Nova Pack, 4µm, 3.9X150 mm) at 40°C eluted with a mobile phase of methanol:water (6:94 v/v) at a flow rate of 2 mL/min. UV detection was carried out at 254 nm. Retention times were 1.9 minutes for iohexol and 5.2 minutes for the IS. Extraction recovery of iohexol in serum was 99.0-104.2% and the calibration curve was linear (r= 0.999) in a concentration range from 10 to 1000 μg/mL (n=6).
