The drug absorption process after an oral administration mainly depends on the drug release from the dosage form, described by its dissolution kinetics, as well as the drug's solubility and permeability under physiological conditions. Nowadays, there is a practical need to improve the evaluation of oral drug bioavailability and it has been proposed that the drug intestinal absorption could be estimated through the compound's permeability determined in models which resemble the absorption process. Thus, the characteristics of the intestinal epithelium and the mechanism of drug transport are described. Different models to estimate intestinal absorption are discussed, their characteristics and predicting capabilities.
