In vitro germination and micropropagation techniques allow to rescue and propagate plants on a large scale. The present work evaluated different cultivation treatments to determine the optimal micropropagation protocol. It is possible to establish an in vitro protocol for optimal germination and cloning of Heimia salicifolia based on the requirements for germination and propagation in situ.Germination protocol was applied at different concentrations of cytokinins and gibberellins in MS medium (1962) at 100% with 3% sucrose (Table 1), but the variables obtained by the different treatments allow to conclude that the best method for germination was without vitamins reducing the cost and obtaining the same results. The effect of these growth regulators on germination time and plant development for four months was evaluated according to the number of leaves, length in centimeters and vigor of seedlings. There were significant differences between the treatments in all the variables, concluding that the best treatment for germination and maintenance of vitroplants is without hormones.For micropropagation protocol was necessary the evaluation of 100% MS medium with different concentrations of 2ip, BA and Kinetin, supplemented with 30 g L-1 of sucrose, 100 mg L-1 of Myo-inositol, 0.40 mg of Thiamine, 60 mg L-1 of L-cysteine and pH of 5.7 ± 0.1. (Table 3). There were significant differences between basal and apical explants. Treatment 2 (0.3 mg L-1 2ip, BA and Kinetin) showed the best results in terms of number of shoots and number of leaves. However, the best results in longitudinal growth of the seedlings were obtained with the treatment without hormones. These two treatments showed to be effective for micropropagation allowing to conclude that it is possible to establish a micropropagation protocol for Heimia salicifolia from apical and basal explants with exponential growth.
