Abstract

The production of laccase from the ligninolytic fungi Coriolopsis gallica was carried out at pilot plant scale in a 100 L bioreactor. A cheap fermentation medium was used containing 3% Bran Flakes in 60 mM phosphate buffer, pH 6.0 containing 0.8 mM copper sulfate. A total of 630 650 laccase units were obtained in 7-days cultures. After few purification steps, including centrifugation, ultracentrifugation, ammonium sulfate precipitation, and two ionic exchange chromatographies, a partially purified preparation was obtained with 79% of laccase showing a specific activity of 313 U/mg.
